Protective Effects of Methanol Extract from Acanthopanacis cortex Shoots against Photoaging in Human Skin Fibroblast

Abstract

Exposure to ultraviolet (UV) light can cause skin photoaging, which is characterized by thickening, wrinkling, pigmentation, and dryness. Ultraviolet exposure is also closely associated with down-regulation of collagen synthesis and up-regulation of matrix metalloproteinases (MMPs). In this study, we investigated the protective effects of methanol extract from Acanthopanacis cortex shoots (ACS) against UVB-induced photoaging in human skin fibroblasts (Hs68 cells). The ACS extract did not show any cytotoxicity at levels up to 50 ¥ìg/mL. The extract showed protective effects against UVB-induced (30 mJ/cm2) cell damage in a dose dependent manner. In addition, the extract significantly reduced UVB-induced generation of reactive oxygen species by 11, 18.2, and 31.6% at concentrations of 12.5, 25, and 50 ¥ìg/mL, respectively, when compared to UVB-irradiation alone. The extract prevented decreases in collagen levels by inhibiting the increases in MMP-1 and MMP-3 levels caused by exposure to UVB-irradiation in Hs68 cells. Taken together, these data indicate that methanol extract from ACS exerts anti-photoaging activities and therefore has the potential for use in cosmeceutical preparations.
